To examine the possible role of PLD in NaCl-stress-induced microtubule reorganization, we compared root growth of wild-type Arabidopsis thaliana, Col-0 with an Arabidopsis thaliana, Col-0 T-DNA insertion mutant line Atpldδ as well as with Arabidopsis plants stably transformed with 35S::GFP-PLDδ. To evaluate the phenotype during normal growth and development, we first grew seedlings on normal media for five days, after five days seedling were transferred at new plates with or without salt (150 or 200 mM NaCl). We have monitored changes in root growth on daily basis for the next six days. The resulting square-root transformed data were then fitted by linear mixed-effects model to evaluate statistical significance of trends across interacting variables. In result growth of pldδ mutant was about 0.36+-0.07 or 0.49+-0.10 mm/day slower when growing on 150 or 200 mM NaCl respectively, with highly significant difference (p = 9.409e-07 ***; Fig. 5 and Fig S3.). While the difference in inhibition of root elongation was especially pronounced in the 150mM treatment for pldδ, in this treatment the overexpressing line GFP-PLDδ have also shown difference in root growth rate, it was growing faster than control with statistical significance (p = 0.0283 *; Fig. 5b). In conclusion, we have observed changes in root growth rate caused by changes in microtubules dynamic which are more severe when PLDδ is lacking.
