Class knowledge-

 The Bradford Method is an analytical procedure that determines the concentration of protein within a sample.

Dye used: “Coomassie Brilliant Blue G-250” which forms a non-covalent complex with proteins.

How does the dye interact: Through electrostatic and Van der Waals interactions, the dye causes a disruption within the protein.

1.Electrostatic interactions occur between ionizable groups, like amino and carboxyl groups of the proteins to the electrons from the Dye.

2.non-covalent interactions allow the dye to be able to interact with the protein. 

Absorption changes-

Once the dye and protein interact, the absorption maximum changes from 495nm to 595nm. The absorptivity change is indicative of a successful interaction.

Pros and cons of the bradford method-

1.Bradford method has many advantages and one of them is that it can be used on certain proteins that have been reduced by reducing agents, which separates it from other methods like Lowry protein assay.

2.Drawbacks to Bradford include its incompatibility with detergents, which at times can be used to solubilize membrane proteins.

Regarding reducing agents:

Reducing agents like Dithiothreitol (DTT), B-Mercaptoethanol are routinely used in protein samples. You can remove these interfering substance through precipitation (by adding acetone or TCA)

Protein samples usually contains salts, solvents, buffers, preservatives, reducing and metal chelating agents. These molecules are frequently used for solubilizing and stabilizing proteins. Other protein assay like BCA and Lowry are ineffective because molecules like reducing agents interfere with the assay. Using Bradford can be advantageous against these molecules because they are compatible to each other and will not interfere.

On March 21, 1937, a peaceful march was organized by the Nationalist Party to commemorate the ending of slavery in Puerto Rico in 1873 by the governing Spanish National Assembly. The police, under the orders of General Blanton Winship, the US-appointed colonial Governor of Puerto Rico, opened fire at the peaceful Puerto Rican Nationalist Party parade, bringing about what came to be known as the "Ponce massacre": 19 people (including two policemen) were killed and over 100 were wounded.[55]

Group Contribution.

Both Sal and Abdullah fact checked the statistics regarding the death toll of the Ponce Massacre and accuracy of the wounded and individuals involved in the massacre. Sal also assisted in adding citations and resources into the article when we corrected it.

Sonia searched information regarding the Spanish National Assembly but found no concrete source regarding the leader of the Spanish National Assembly or who ended slavery in Puerto Rico. She also researched about the dates these significant events occurred and focused on editing the article, checked for grammar and punctuation.
