Was Ai the only chimpanzee initially involved in the Ai project?

It's not true. According to Primate Research Institute (1980) or Matsuzawa (2003), it was Ai, Akira, and Mari who were initially involved in the Ai project. The appendix II in Primate Research Institute (1980) tells us in details what was conducted on what day in the series of experiments.
