English: A simplified explanation of how tagged proteins can be utilized in performing a pull-down assay. This technique is useful for determining the existence of any protein-protein interaction between tagged and purified "bait" and "prey" proteins (chosen by the experimenter). Although the S-tag and FLAG-tag are featured here, different tags can be used in a similar assay.
