Spruce Street is notable as one of William Penn's original streets in his plan for building the "1200 acre site of Philadelphia." Spruce Street is one of the original in Penn's plan that are named after a tree traversing east-west, termed an "odanym".

There was an anecdote written by John Fanning Watson who told a story of "water in basements" in houses along Spruce Street, which he said may have pointed to when the street was a water bog, which may have had water vessels sailing on it.

At 8th Street and Spruce is the historic Pennsylvania Hospital, which was founded by Benjamin Franklin. Between 11th and 12th Street, Spruce Street is also notable for the LGBT community.
