Summary

Description
English: The Adapter sequences AD1L and AD1R are ligated to the size selected DNA. The adapters complement each other and the DNA is circularized. A restriction enzyme is added that cleaves the DNA. AD2L and AD2R are ligated, the DNA is circularized and a restriction enzyme cleaves the DNA. AD3L and AD3R are added, the DNA is circularized, and a type III restriction enzyme is added. This cleaves out a 300 - 400bp piece of DNA and AD4 replaces it to complete the circular, adapter ligated template DNA.
